Recent studies have provided mostly indirect evidence for oxygen-depleted zones inside seed structures. In this study, optical sensors were used to analyze detailed oxygen profiles across developing seeds. They showed that oxygen concentration decreases sharply to about 1% within the seed coat. This suggests that oxygen entry into the seed is strongly restricted by the cutinized cell layers covering the seed, and that seeds develop in an hypoxic environment. Oxygen concentration increased upon illumination, indicating that photosynthesis significantly contributes to internal oxygen levels in these green seeds. Although oxygen concentration and energy state fell to low levels, anoxic zones were not detected within seeds.

Using a set of assays that were based on a novel enzymic-cycling system, metabolite levels in response to a stepwise decrease in external oxygen concentration were determined in Arabidopsis seeds.

The expression profile of Arabidopsis root cultures in low oxygen included not only genes encoding the known anaerobic proteins but also several transcription factors, signal transduction components, and genes that encode enzymes of pathways not previously known to be involved in low-oxygen metabolism. The regulatory regions of genes with a similar expression profile contained similar sequence motifs, suggesting the coordinated transcriptional control of groups of genes by common sets of regulatory factors.

Overexpression of the non-symbiotic Arabidopsis GLB1 or the symbiotic GLB1S from Parasponia andersonii in transgenic Arabidopsis plants leads to increased growth under normal oxygen conditions and increased survival under severe hypoxia. Hypoxia tolerance was lost when a mutated GLB1 with low oxygen affinity was overexpressed, demonstrating that a high ligand-binding affinity is essential for increased tolerance of hypoxic stress. As the oxygen-binding affinity of GLB1 is greater than that of cytochrome oxidase, it is unlikely that GLB1 assists the transport of oxygen into mitochondria.
