Plant J 2002, 29:427-437. The hys1 mutant is isolated as an accelerated-leaf-senescence mutant. It is revealed to be allelic to the cpr5 mutant, which has a constitutive defence-response phenotype. Furthermore, this mutant is hyper-responsive to exogenously supplied sugars at the seedling stage. The HYS1 gene is cloned and found to encode a novel membrane protein.

T-DNA-based cloning revealed that ORE4-1 encodes plastid ribosomal protein small subunit 17. The authors also show the decreased activity of photosystem I in ore4-1 mutants.

The ore9 mutant showed a delayed-leaf-senescence phenotype during age-, dark- and hormone-induced senescence. The ORE9 gene encodes an F-box protein and can interact with ASK1, a component of the plant SCF complex, indicating that the SCF complex has a role in leaf senescence.

The corresponding gene encodes R-transferase, a component of the N-end rule proteolysis pathway. A transient expression assay shows that the dis1 mutant has deficiency in N-end-rule-dependent proteolysis.

The authors identified an acyl hydrolase gene, SAG101, that is upregulated during senescence. Leaf senescence is delayed in transgenic plants in which SAG101 is repressed, and overexpression of this gene can lead to premature senescence. These results suggest that lipid degradation plays a regulatory role in leaf senescence.

The authors describe how they used cDNA AFLP analysis to search for targets of the senescence- and defence-associated transcriptional factor AtWRKYC. Several signal-transduction components were identified as possible targets of AtWRKY6. One of these targets, a novel SIRK is especially active in senescing leaves.
