This is a very good study showing that hepatic lipase enhances VLDL degradation via LDL receptor pathway by concentrating VLDL particles onto cell surface prior to receptor interaction. One of a few studies showing that hepatic lipase binds to LDL receptor and enhances VLDL catabolism.

One of many papers from this excellent group showing that hepatic lipase and scavenger receptor SR-B1 expression in kidney cells results in selective uptake of HDL cholesterol esters. This paper shows that inactive mutant hepatic lipase also enhances direct HDL cholesterol ester uptake.

Biochemistry 1998; 37:14974-14980. This is the first study that shows that HDL remodelling by hepatic lipase modifies binding and internalization of triglyceride rich HDL into hepatoma cells. This paper suggests that liver uptake of remodelled remnant HDL occurs after hepatic lipase action. This may explain kinetic studies showing that apoHDL is taken up by the liver.

A transgenic rabbit study showing that hepatic lipase and apolipoprotein E expression reduced diet induced elevation in plasma cholesterol, small VLDL, IDL and HDL cholesterol. Effects of hepatic lipase and apolipoprotein E were synergistic. This provides evidence on the role of hepatic lipase in HDL metabolism and supports in vitro evidence on the interaction of hepatic lipase and apolipoprotein E.

This is one of many excellent transgenic mice studies from this group. Double knockout mice, hepatic lipase and apolipoprotein A-II deficient had higher HDL cholesterol, increased HDL size and decreased HDL cholesterol ester fractional catabolic rate when compared to apolipoprotein A-II knockout mice. Apolipoprotein A-II knockout mice showed active HDL triglyceride lipolysis by hepatic lipase. These studies suggested that apolipoprotein A-II is a physiological inhibitor for hepatic lipase and suggests that HDL A-I only particles are metabolically more active than HDL A-I/A-II particles.

Circulation 1998; 98(Suppl):1-452.[abstract] This study showed that lower hepatic lipase activity was associated with elevated levels of larger HDL particles and decreased levels of smaller HDL particles. It suggested that large HDL particles indeed are substrate particles for hepatic lipase and that hepatic lipase may be involved in conversion of larger HDL particles into smaller HDL particles.
