Hepatic triglyceride lipase promotes low density lipoprotein receptor-mediated catabolism of very low density lipoprotein in vitro. J Lipid Res 1999; 40:1263-1275. Remnant high density lipoprotein2 particles produced by hepatic lipase display high-affinity binding and increased endocytosis into a human hepatoma cell line (HepG2). Biochemistry 1998; 37:14974-14980. This is the first study that shows that HDL remodelling by hepatic lipase modifies binding and internalization of triglyceride rich HDL into hepatoma cells. This paper suggests that liver uptake of remodelled remnant HDL occurs after hepatic lipase action. This may explain kinetic studies showing that apoHDL is taken up by the liver.

Hepatic lipase facilitates the selective uptake of cholesteryl esters from remnant lipoproteins in apoE-deficient mice. J Lipid Res 1998; 39:2150-2160. An excellent study showing that catalytically inactive hepatic lipase mutant in transgenic mice enhances selective uptake of cholesterol esters. Provides evidence that hepatic lipase may act as a ligand in an animal model. This action of hepatic lipase is independent of lipolysis.

Overexpression of human hepatic lipase and ApoE in transgenic rabbits attenuates response to dietary cholesterol and alters lipoprotein subclass distributions. Arterioscler Thromb Vasc Biol 1999; 19:625-632. A transgenic rabbit study showing that hepatic lipase and apolipoprotein E expression reduced diet induced elevation in plasma cholesterol, small VLDL, IDL and HDL cholesterol. Effects of hepatic lipase and apolipoprotein E were synergistic. This provides evidence on the role of hepatic lipase in HDL metabolism and supports in vitro evidence on the interaction of hepatic lipase and apolipoprotein E.

    • 40 Weng W, Brandenburg NA, Zhong SB, Halkias J, Wu L, Jiang XC, et al. ApoA-II maintains HDL levels in part by inhibition of hepatic lipase: studies in apoA-II and hepatic lipase double knockout mice. J Lipid Res 1999; 40:1064-1070. This is one of many excellent transgenic mice studies from this group. Double knockout mice, hepatic lipase and apolipoprotein A-II deficient had higher HDL cholesterol, increased HDL size and decreased HDL cholesterol ester fractional catabolic rate when compared to apolipoprotein A-II knockout mice. Apolipoprotein A-II knockout mice showed active HDL triglyceride lipolysis by hepatic lipase. These studies suggested that apolipoprotein A-II is a physiological inhibitor for hepatic lipase and suggests that HDL A-I only particles are metabolically more active than HDL A-I/A-II particles.

    • Effect of estrogen on hepatic lipase and HDL particle size in post menopausal women
    • 50 Thuren TY, Rudel LL, Brinton EA, Herrington DM. Effect of estrogen on hepatic lipase and HDL particle size in post menopausal women. Circulation 1998; 98(Suppl):1-452.[abstract] This study showed that lower hepatic lipase activity was associated with elevated levels of larger HDL particles and decreased levels of smaller HDL particles. It suggested that large HDL particles indeed are substrate particles for hepatic lipase and that hepatic lipase may be involved in conversion of larger HDL particles into smaller HDL particles.
