    • The authors investigate the role of the usher in pilus biogenesis. They demonstrate that the chaperone-adhesin complex binds most rapidly and tightly to the usher. The chaperone-adhesin-usher ternary complex was purified and the usher shown to have adopted a trypsin-resistant conformation in the complex that is maintained during pilus assembly. A model in which binding of the chaperone-adhesin complex stabilizes the usher and initiates pilus assembly is suggested.

    • Type IV pili, transient bacterial aggregates, and virulence of enteropathogenic Escherichia coli
    • The authors demonstrate that functional type IV bundle-forming pili of enteropathogenic E. coli (EPEC) are required for maximum virulence in humans. Mutant strains lacking a functional type IV pilin (bfpA) or transcriptional activator (bpfT) gene caused much less diarrhea than did a wild-type strain. Wild-type EPEC forms autoaggregates that disperse over time; a mutation in bfpF, thought to provide energy for pilus biogenesis, produces a strain that forms morphologically distinct autoaggregates that fail to disperse over time. This mutant is more adherent than wild-type EPEC, and forms pedestals and causes actin condensation as does wild-type EPEC; however, the mutant is less virulent than wild-type EPEC, suggesting that bacterial dispersal after aggregation is important for virulence in humans.
