The anion channel was inhibited by 4-acetamido-4′-isothiocyano-2,2′ disulfonic acid stilbene, the glutathione system by N-ethylmaleimide, and the catalase system by aminotriazole. An intact glutathione system was necessary for the protective effect, but catalase was not. This analysis of the mechanism of projected erythrocyte protection provided empirical evidence for the observations and speculations of previous investigators on the usefulness of a system of erythrocyte-based scavenging of oxygen free radicals.

The methodology for tracking the aging erythrocytes relied on analysis of biotin-magnetic bead-bound, fluorescently labeled avidin cell-sorted, biotinylated cells from two dogs in which 97% to 100% of the circulating cells were labeled. The cells recovered at each time point during the 119-day cycle thus represented a progression from an approximate random age mix (day 1) to an almost pure age cohort (day 119). The value of such an approach lies in the ability to analyze the natural oxidative changes occurring with advancing age.

The underlying hypothesis was that if there were any normalizing effect of the skeleton on maintaining asymmetry, then inhibition of the flipase would result in greater PS externalization in such membrane-skeleton uncoupled cells, relative to that induced in normal erythrocytes under the same conditions. However, inhibition of the flipase by either sulfhydryl modification or rapid ATP depletion did not result in PS externalization above the low levels produced under the same conditions in normal erythrocytes, even allowing for slow migration of PS to the outer monolayer by overnight incubation at room temperature. This is a clear empirical demonstration that the flipase is not regulated by any component of the membrane skeleton.
