Isotype control antibodies were rat immunoglobulin 2a (lgG2a) and rat lgG2b (Pharmingen).

B6.2.16 cells were harvested and stained with T3.70-biotin (followed by streptavidin-PE) and anti-CD3(2C11)-CyC (Pharmingen), fixed in 1% paraformaldehyde, permeabilized with 0.03% saponin, and stained with mAb to perform [A. E. A. Kawasaki, Int. Immunol. 7, 677 (1990)] or isotype control (rat IgG2a, Pharmingen) in 0.3% saponin with 20 μl of normal goat serum (Accurate, Westbury, NY). The stained cells were washed with 0.03% saponin and stained again with a goat anti-rat IgG-PE secondary antibody (Southern Biotechnology Associates, Birmingham, AL). The stained samples were analyzed by FACS for perform expression level above that of the isotype control.
