10. + cells was quantitated by staining with anti-CD8 conjugated to CyChrome (CyC) then analyzed by FACS for surface molecule expression and CFSE green fluorescence intensity (cell division). Antibodies used for FACS analysis were anti-CD8-CyC and anti-CD44-phycoerythrin (PE) (Pharmingen, San Diego, CA). Isotype control antibodies were rat immunoglobulin 2a (lgG2a) and rat lgG2b (Pharmingen).

After 4 hours at 37°C, the degree of cytolysis was determined and the percentage specific lysis calculated as follows: [(experimental release - spontaneous release)/(maximum release - spontaneous release)] × 100. The percentage specific lysis of the targets in the absence of H-Y peptide was never more than 3% at the maximum E:T ratio of 30:1, and the level of spontaneous lysis never exceeded more than 6% of maximal lysis.

B6.2.16 cells were harvested and stained with T3.70-biotin (followed by streptavidin-PE) and anti-CD3(2C11)-CyC (Pharmingen), fixed in 1% paraformaldehyde, permeabilized with 0.03% saponin, and stained with mAb to perform [A. E. A. Kawasaki, Int. Immunol. 7, 677 (1990)] or isotype control (rat IgG2a, Pharmingen) in 0.3% saponin with 20 μl of normal goat serum (Accurate, Westbury, NY). The stained cells were washed with 0.03% saponin and stained again with a goat anti-rat IgG-PE secondary antibody (Southern Biotechnology Associates, Birmingham, AL). The stained samples were analyzed by FACS for perform expression level above that of the isotype control.
