The authors perform an extensive transcriptional analysis using in situ hybridization in vitro and in vivo, and find that the M3 region of the γHV68 genome is transcribed in infected spleen cells at times when many probes for other genes are negative. This argues for an interesting role for M3 in γHV68 infection, perhaps during the transition to latency.

The authors demonstrate that B-cell-deficient mice maintain a high frequency of cells that reactivate latent γHV68 at times post-infection when normal mice are transitioning from having a high frequency of cells that reactivate the virus to a low frequency. In addition, this paper presents the first assay for quantifying the frequency of cells carrying γHV68 genome during latency. This analysis demonstrated that B cells regulate the efficiency of reactivation from latency rather than the frequency of cells carrying γHV68 genome.

The authors show that macrophages carry latent γHV68, and confirm that B cells carry latent γHV68 genome during latency. This is an important finding as it demonstrates that there are multiple cellular reservoirs of latent γHV68. In addition, they show that latent cells can be recruited into sites of inflammation, arguing that a reservoir of cells in the bone marrow may supply latently infected cells to tissues.

J Virol 1999, 73:2321-2332. This is the first paper to clearly demonstrate transcription of the γHV68 genome in latent tissues. Several candidate latency genes are identified. Interestingly, several of these are shared with the human and primate herpesviruses Epstein-Barr virus, herpesvirus saimiri and Kaposi's sarcomaassociated herpesvirus. This argues that γHV68 latency will share important molecular mechanisms of latency with primate γ-herpesviruses.

Transgenic studies showed that the v-cyclin promotes cell cycle progression in thymocytes, and induces lymphoblastic lymphoma in a significant fraction of transgenic mice. This identifies the v-cyclin as the first γHV68 oncogene.
