Evidence suggest that a CBR-ligand system exists in the immune system and has some role in immune homeostasis and that cannabinoids modulate immunity. The review also emphasizes that the function and distribution of receptor subtypes in various immune compartments, and many other basic issues, still need clarification.

    • 9 tetrahydrocannabinol are neuroprotective antioxidants
    • 9 tetrahydrocannabinol are neuroprotective antioxidants. Proc Natl Acad Sci USA 1998, 95:8268-8273. The antioxidant capacity, proven by cyclic voltametry and in a chemical (Fenton reaction) system for THC and cannabidiol (and earlier for the nonpsychotropic synthetic cannabinoid HU-211) appears to be a common property of classical cannabinoid structures. Obviously, the phenol is responsible for this property. The potential use of nontoxic cannabidiol as a neuroprotective agent (e.g. in ischemic stroke) is of particular interest.

Life Sci 1998, 63:PL1-6. THC does not induce the maximal effect obtained by classical CBR agonists such as 8; in fact, at high concentrations, THC exhibits antagonistic properties. Cannabinol is a weak agonist and cannabidiol is an antagonist acting in the micromolar range. The variable ratio of THC, cannabinol and cannabidiol in marijuana might influence its pharmacological effects.

    • Control of pain initiation by endogenous cannabinoids
    • Calignano A, La Rana G, Giuffrida A, Piomelli D: Control of pain initiation by endogenous cannabinoids. Nature 1998, 394:277-281. Palmytylethanolamide (PEA) (23) exerts similar effects (i.e. control of pain initiation) by activating peripheral CB2-like receptors. Administered together, AEA and PEA act synergistically, reducing pain responses 100-fold more potently than each compound alone. Since 10 (a CB1 antagonist) and 16 (a CB2 antagonist) prolong and enhance the pain behavior, it is obvious that peripheral CB-like receptors participate in the intrinsic control of pain initiation and that locally generated AEA and PEA may mediate this effect.
