-
Liu R, Barrick J, Szostak JW, Roberts RW: Optimized synthesis of RNA-protein fusions for in vitro protein selection. Methods Enzymol 2000, in press. This paper details significant improvements in yield of mRNA-protein fusion synthesis. In addition, it is the first to demonstrate that a full-sized protein can be generated as a fusion, and that RNA binding peptides retain specificity as naked fusions.

-
Hanes J, Plückthun A: In vitro selection and evolution of functional proteins by using ribosome display. Proc Natl Acad Sci USA 1997, 94:4937-4942. The first report that a single-chain antibody's protein could be generated and screened by bacterial ribosome display. Extensive optimization and assays are delineated to assure protein production and mRNA stability; however, many of the clones assayed do not bind as well as the wild type.

Ribosome display efficiently selects and evolves high-affinity antibodies in vitro from immune libraries
-
Hanes J, Jermutus L, Weber-Bornhauser S, Bosshard HR, Pluckthun A: Ribosome display efficiently selects and evolves high-affinity antibodies in vitro from immune libraries. Proc Natl Acad Sci USA 1998, 95:14130-14135. This is the first report of construction and implementation of an immunized murine single-chain antibody library using ribosome display. Here, the best clone appears to have 65-fold higher affinity than the sequence it was derived from. Mutations were either incorporated in the primers or occured during the selection cycles.
