The authors show that this promoter depends on Pax6- and Maf-binding sites. A short promoter region containing these sites, however, displayed the activity not only in the lens but also in the brain of transgenic mice, and an additional upstream region was required for suppression of the activity in the brain. This paper illustrates a good example of how interplay of transcription factors leads to a lens-specific expression pattern.

We first show that Sox1-3 are essential transcription factors for activation of δ-crystallin enhancer and demonstrate, using chicken embryo, that expression of Sox2/3 in the presumptive lens area of head ectoderm is activated by the inductive influence of the optic vesicle, and Sox1 is activated just before lens fiber differentiation begins. In the mouse, Sox3 is not appreciably expressed in the lens and Sox2 expression is turned off in the lens fiber when Sox1 is activated. The results indicate that Sox2/3 activation is fundamental to the process of lens induction.

We show that action of Sox proteins requires interaction with specific partner factors bound at a nearby site of the enhancer. This is the mechanism of the specific action of the Sox proteins on individual regulatory sites.

This paper reports a clear-cut knockout mouse analysis of the Sox1 function. The major phenotype is the defect in development of the lens fiber where Sox1 functions as major Sox. All γ-crystallin gene expression is eventually lost and fiber cell elongation is arrested, indicating that these genes and the process are under strict regulation by Sox1 in the mouse.

BMP4 mutant mice die around the time of lens induction but the authors have here taken advantage of organ culture and show that Sox2 is not induced in the head ectoderm and lens induction does not take place in the mutant eye. Addition of BMP4 to the retina-ectoderm conjugant culture rescues Sox2 expression and lens development in the mutant tissue, whereas addition of BMP4 to the ectoderm alone does not elicit lens development. The results demonstrate that BMP4 is essential for Sox2 induction and initiation of lens differentiation but that BMP4 per se is not an inducer of Sox2.

Through analysis of a set of appropriate molecular markers, they demonstrate that lens-fiber differentiation is initiated, as indicated by synthesis of most fiber-specific crystallins and intermediate filament components, but is arrested at its initial stage failing to turn off of E-cadherin or to activate cyclin-dependent kinase inhibitors. Prox1 is thus essential for transition from lens epithelium to lens fiber.
