    • Activation of gene expression by a novel DNA structural transmission mechanism that requires supercoiling-induced DNA duplex destabilizatlon in an upstream activating sequence
    • G promoter in a region that is required for IHF activation. Similar threshold superhelical densities are required for duplex destabilization and promoter activation. Binding of IHF prevents duplex destabilization with concomitant destabilization at -10 suggesting a supercoil-dependent DNA structural transmission mechanism.

Activation by an A-tract-containing sequence placed upstream of core promoters requires the carboxy-terminal domain of the RNAP α subunit. Activation is optimal when the A-tract is present close to the -35 hexamer. These characteristics suggest that A-tracts function as UP elements and do not activate through bending. This paper provides a mechanistic explanation for A-tract activation.
