Immunohistochemical demonstration of enzymatically modified human LDL and its colocalization with the terminal complement complex in the early atherosclerotic lesion

Torzewski M, Klouche M, Hock J, Messner M, Dorweiler B, Torzewski J, et al: Immunohistochemical demonstration of enzymatically modified human LDL and its colocalization with the terminal complement complex in the early atherosclerotic lesion. Arterioscler Thromb Vasc Biol 1998, 18:369-378. This article suggests a new pathogenetic mechanism that could explain how modified lipoproteins might affect the immune system and especially how lipoproteins sequestered in the intima could activate the complement system.

Ridker PM, Cushman M, Stampfer MJ, Tracy RP, Hennekens CH: Inflammation, aspirin, and the risk of cardiovascular disease in apparently healthy men. N Engl J Med 1997, 336:973-979. This landmark paper provides evidence that increased levels of C-reactive protein are a significant risk factor for myocardial infarction and raises a question that the effect of an aspirin regimen could be the result of decreased inflammation.

Hypercholesterolemia is associated with a T helper (Th) 1/Th2 switch of the autoimmune response in atherosclerotic apoE-knockout mice
-
Zhou X, Paulsson G, Stemme S, Hansson GK: Hypercholesterolemia is associated with a T helper (Th) 1/Th2 switch of the autoimmune response in atherosclerotic apoE-knockout mice. J Clin Invest 1998, 101:1717-1725. This detailed study provides evidence for T-cell help with production of Th1-dependent IgG2a antibodies against oxidized LDL that peak during the earliest phase of atherosclerotic lesion and with interferon-γ both in the spleen and in the plaque itself Moreover, it shows that severe hypercholesterolemia is associated with a switch toward a Th2-driven response, with interleukin-4 predominating in the atherosclerotic lesions.

High anticytomegalovirus (CMV) IgG antibody titer is associated with coronary artery disease and may predict post-coronary balloon angioplasty restenosis
-
Blum A, Giladi M, Weinberg M, Kaplan G, Pasternack H, Laniado S, Miller H. High anticytomegalovirus (CMV) IgG antibody titer is associated with coronary artery disease and may predict post-coronary balloon angioplasty restenosis. Am J Cardiol 1998, 81:866-868. This prospective study demonstrates an association between high anti-CMV antibody titer and restenosis after coronary balloon angioplasty. The results suggest that a chronic immunologic response may play a role in atherosclerosis and restenosis.

Infection with Chlamydia pneumoniae accelerates the development of atherosclerosis and treatment with azithromycin prevents it in a rabbit model

Circulation 1998, 97:633-636. Using a chronic animal model, the authors demonstrate that C. pneumoniae infection leads to atherosclerotic lesions and that development of these lesions could be prevented by antibiotic regimens.

Inflammation, pravastatin, and the risk of coronary events after myocardial infarction in patients with average cholesterol levels
-
Ridker PM, Rifai N, Pfeffer MA, Sacks FM, Moye LA, Goldman S, et al. for the Cholesterol and Recurrent Events (CARE) Investigators: Inflammation, pravastatin, and the risk of coronary events after myocardial infarction in patients with average cholesterol levels. Circulation 1998, 98:839-844. A nested case-control study of a large secondary prevention trial demonstrates that plasma levels of C-reactive protein and serum amyloid A predict the risk of recurrent coronary events among stable patients with a history of prior myocardial infarction. The data also raise the possibility that the effect of inflammation on coronary risk may be attenuated by pravastatin therapy.

Visceral abdominal-fat accumulation associated with use of indinavir
-
Miller MD, Jones E, Yanovski JA, Shankar R, Feuerstein I, Falloon J. Visceral abdominal-fat accumulation associated with use of indinavir. Lancet 1998, 351:871-875. This article reports on unusual side effects of antiviral therapy in AIDS patients. The pattern of body fat redistribution resembles that of the metabolic syndrome regarded as a major risk factor for coronary atherosclerosis.
