Blood 1998, 91:222-230. GM-CSF tumor cells were capable of rejecting a large tumor burden as compared with tumor cells transduced with other genes. Interestingly, other cells in addition to T cells seemed to mediate the specific tumor-killing effect. This finding might have great implications for the future development of tumor vaccination strategies.

    • 40 Shetye J, Ragnhammar P, Liljefors M, Christensson B, Frödin J-E, Biberfoldt P, Mellstedt H: Immunopathology of metastases in patients of colorectal carcinoma treated with monoclonal antibody 17-1A and granulocyte macrophage colony-stimulating factor. Clin Cancer Res 1998, 4:1921-1929. Analyzing effector functions at the tumor site during immunotherapy should be one of the most appropriate approaches to the study of mechanisms of importance. This study shows that relevant effector cells containing cytotoxic granulae infiltrated the tumor. An antitumor T-cell response seemed to be initiated with the induction of cytotoxic CD8 and CD4 T cells, probably as a result of an idiotypic network response. CD8 and CD4 T cells showed a differential display in the tumor.

In vivo primed GM-CSF cells clearly produced T- and NK cell-suppressive factors. This information is of great importance for the utilization of GM-CSF as an immune-augmenting CSF.

Treatment with G-CSF-mobilized peripheral blood stem cells reduces the severity of graft-versus-host disease. This might be explained by hyporeactive T cells. A reversible defect in T-cell signaling by a secreted monocyte product might be responsible. This effect may add positively to the stem cell-mobilizing effect of G-CSF in allogenic stem cell transplantation.
