    • Rapid actin-based plasticity in dendritic spines
    • Using hippocampal neurons transfected with GFP-labelled actin, the authors showed that dendritic spines undergo spontaneous, actin-dependent shape changes. The changes are rapid, occurring on a time-scale of seconds, and involve spines that are contacted by axon terminals. These results demonstrate that spine plasticity is not just a developmental phenomenon, but that it persists at established synapses

    • Change in the shape of dendritic spines caused by overexpression of drebrin in cultured cortical neurons
    • Using GFP-tagging, the authors show that drebrin, an actin-binding protein, is targeted to dendritic spines through its actin-binding domain. Overexpression of drebrin induces filopodia on normally smooth epithelial cells and has a dramatic effect on dendritic spines, increasing their average length by up to 50% without changing their number. This is one of several recent papers testifying to the large number of proteins that influence the function of postsynaptic actin

    • Using hippocampal slice cultures, the authors show that either lesioning axons or blocking postsynaptic AMPA receptors leads to extensive loss of dendritic spines and shrinkage of those that remain. These effects were not produced by blocking NMDA receptors. Conversely, the effect of lesioning could be rescued by supplying cultures with small quantities of AMPA. Using hippocampal slice cultures, the authors show that either lesioning axons or blocking postsynaptic AMPA receptors leads to extensive loss of dendritic spines and shrinkage of those that remain. These effects were not produced by blocking NMDA receptors. Conversely, the effect of lesioning could be rescued by supplying cultures with small quantities of AMPA. The results implicate subthreshold AMPA receptor activation, produced by spontaneous quantal neurotransmitter release, in spine maintenance.
