    • Claudin-1 and -2: Novel integral membrane proteins localizing at tight junctions with no sequence similarity to occludin
    • The authors report identification and cloning of genes encoding claudin-1 and claudin-2. Claudin-1 and claudin-2 were identified from isolated junctional fraction from the liver as proteins that co-partition with occludin during sonication and sucrose density gradient centrifugation. FLAG-tagged claudin-1 and claudin-2 were targeted to and incorporated into pre-existing TJ strands, when introduced into cultured MDCK cells.

    • A single gene product, claudin-1 or -2, reconstitutes tight junction strands and recruits occludin in fibroblasts
    • This is the first report that TJ strands can be reconstituted within plasma membranes of cultured fibroblasts by a single gene product, claudin-1 or claudin-2. Importantly, it also shows that occludin is recruited to and integrated into the claudin-based backbone of reconstituted TJ strands.

Although information regarding this molecule is still limited, this is the first molecule bearing a single transmembrane domain shown to be concentrated in the TJ region of epithelial and endothelial cells.

    • This is an important report showing that a mammalian homologue of PAR-3, C. elegans polarity gene product, is exclusively concentrated at the cytoplasmic surface of TJs in simple epithelial cells. This is the first description of the close relationship between polarisation of mammalian epithelial cells and determination of the cell division axis in early embryos of C. elegans
