The knockout strain was transformed with the relevant p177 constructs and grown on LB/Kan plates at 30°C. Dilute cell suspensions were made from the resulting colonies. A 1-μl sample of each cell suspension (6700 to 19,200 cells) was pipetted onto the test plates and incubated at the appropriate temperature for 24 hours.
