of outstanding interest. The mutations in a SIN variant with low cytopathogenicity were mapped and tested individually in wild type background. One nsP2 mutation was shown to be associated with persistent infection although the level of replication was as high as in wild type virus. This mutation was incorporated into a SIN vector for β-galactosidase and shown to increase expression.

When the retrovirus recombinant genes were expressed as RNA in cytoplasm of cells together with retroviral structural proteins and enzymes they were shown to be packaged into transduction competent retrovirus vectors at high yield. Packaging did not involve RNA processing (e.g. splicing), which is the case if the RNA is produced in the cell nucleus.
