Rapid determination of single base mismatch mutations in DNA hybrids by direct electric field control

Sosnowski RG, Tu E, Butler WF, O'Connell JP, Heller MJ: Rapid determination of single base mismatch mutations in DNA hybrids by direct electric field control. Proc Natl Acad Sci USA 1997, 94:1119-1123. The authors describe electrical manipulation of DNA to produce novel arrays and to identify mismatches in hybridised duplexes. This could be easily automated but individual connections to a large array of electrodes could be difficult.

Detection of Heterozygous mutations in BRCA1 using high density oligonucleotide arrays and two-colour fluorescence analysis
-
Hacia JC, Brody LC, Chee MS, Fodor SPA, Collins FS: Detection of Heterozygous mutations in BRCA1 using high density oligonucleotide arrays and two-colour fluorescence analysis. Nat Genet 1996, 14:441-447. This paper offers a useful appraisal of the application of DNA arrays to mutation detection. The 3.45-kb exon 11 of the breast cancer gene, BRCA1 was scanned for heterozygous mutations in fifteen patient and twenty control samples. Normalisation to wild type sequence was done simultaneously using two-colour fluorescence.

Nat Med 1996, 2:753-759. In this paper, oligonucleotide arrays and conventional sequencing have been compared side by side in the determination of variation in the known HIV protease gene sequence. A large degree of natural variation was found, which suggests that different patients are likely to manifest differential response to protease inhibitors.

Quantitative phenotypic analysis of yeast deletion mutants using a highly parallel molecular bar-coding strategy
-
Shoemaker DD, Lashkari DA, Morris D, Mittmann M, Davis RW: Quantitative phenotypic analysis of yeast deletion mutants using a highly parallel molecular bar-coding strategy. Nat Genet 1996, 14:450-456. This important paper describes a method to tag deletion mutants of the yeast Saccharomyces cerevisiae with unique identity sequences that can be quantitatively read by hybridisation to an oligonucleotide array. The method would enable one-pot testing of survival characteristics under different environmental conditions tor all deletion strains.

Milner N, Mir KU, Southern EM: Selecting effective antisense reagents on combinatorial oligonucleotide arrays. Nat Biotechnol 1997, 15:537-541. This paper describes a method for selecting antisense oligonucleotides on scanning arrays and shows correlation between array-selected oligonucleotides against rabbil β-globin and antisense activity in RNAse H and in vitro translation assays. The method should improve the reliability of antisense techniques, which would be useful tools for analysing gene function and for disease therapy.
