The authors show a fundamental difference in protein folding between eukaryotes and prokaryotes. The former fold proteins co-translationally so that for multidomain proteins, frequent amongst eukaryotes, each domain is folded independently. Prokaryotes, on the contary, fold proteins post-translationally and consequently are often unable to fold multidomain eukaryotic proteins properly.

The peptide-binding domain of DnaK, bound to a peptide substrate, was crystallised and the structure was determined at 2.0 Å resolution. The peptide is bound in an extended conformation. The structure suggests the presence of a latch, which presumably keeps the substrate present for a time long enough to permit conformational changes.

The replication initiation protein RepA of plasmid P1 spontaneously forms inactive dimers. These can be monomerised in vitro by the chaperone ClpA, forming active RepA. The reaction involves assembly of CIpA and formation of an unstable complex with RepA dimers, followed by stabilisation of the complex; ATP is required but need not be hydrolysed. Then ATP is hydrolysed and active RepA monomers are released.

In this paper, the authors study the direct interaction in vivo between GroE and a set of proteins and evaluate their residence time. They conclude thai 10-15% of all cytoplasmic protein transits through GroEL.

Gene product 31 of bacteriophage T4 can functionally replace GroES as co-chaperonin with GroEL for protein folding, and its crystal structure, determined at 2.3 Å resolution, resembles that of GroES. Assembly of the 56 kDa capsid protein, however, specifically requires gp31, which increases the size and hydrophilicity of the cavity in which protein folding takes place.

J Biol Chem 1996, 271:27730-27738. The F plasmid proteins CcdA and CcdB form an antidote-poison couple which kills F- (plasmid free) segregants. It is shown here that CcdA is degraded in vitro by Lon protease, in an ATP-dependent reaction, and that CcdB protects CcdA agains degradation. The carboxy-terminal moiety of CcdA, lacking the α helices of intact CcdA, can be degraded by Lon in the absence of ATP, and the same peptide bonds are hydrolysed, showing that ATP hydrolysis is not required for cleavage specificity.

The transposase of phage Mu forms a stable complex with target DNA. Once transposition is completed, this complex is disassembled by the chaperone ClpX, which recognises a 10 amino acid peptide in the carboxy-terminal domain of the transposase. Part of this peptide is also recognised by MuB, which thereby prevents disassembly of transposition intermediates.

This interaction antagonises HflB-dependent degradation of SecY, both in vivo and in vitro.

Cell 1997, 90:415-424. In Caulobacter crescentus the global regulator CtrA governs several cell cycle events: replication initiation, ftsZ expression and flagellar synthesis. CtrA is activated by phosphorylation in mid-S phase. As the cell approaches division, CtrA is degraded specifically in the cell half destined to become the stalk cell but remains intact in the cell half which becomes the swarmer cell, where it rapidly initiates DNA replication.
