Role of the occluding loop in cathepsin B activity
-
of special interest. This study and [24] clarify the structural basis for substrate and inhibitor preferences of cathepsins L and B. The pro-region of both proteins overlies the active-site cleft and prevents substrate entry. In the case of cathepsin L, once the pro-region is removed (mainly by autoactivation at acidic pH) then the cleft is open to substrate entry. In the case of cathepsin B, the occluding loop replaces the pro-region in the cleft of the active enzyme, impairing both substrate entry and inhibition by macromolecular inhibitors such as cystatin C

Li controls the transport of major histocompatibility class II molecules to and from lysosomes
-
of special interest. Prior studies (see [4,7]), have reported the distribution of MHC class II molecules in multiple endosomal/lysosomal compartments. Results of this study of MHC class II trafficking in a murine B-cell line indicates that the efficiency of li processing regulates the distribution of MHC class II within cells. This may be directly relevant to more recent work by the same group showing altered MHC class II trafficking as murine dendritic cells mature to antigen presenters [47]. These findings provide yet another role for proteolytic processing of li in regulation of antigen presentation

Developmental regulation of MHC class II transport in mouse dendritic cells
-
of special interest. This study and [48] show that expression of MHC class II complexes by dendritic cells occurs while the cells are immature and endocytically active but is altered by maturation and exposure of cells to inflammatory stimuli. Mature, stimulated dendritic cells have long-lived MHC classII/peptide complexes on their surfaces. This supports the notion that endocytosed protein may in some way persist in young, migratory dendritic cells only to load and be presented as antigen when the cells mature at sites of T-cell accumulation. It will be important to clarify the protease profile of dendritic cells at these various stages of functional maturation

HLA-DM interactions with intermediates in HLA-DR maturation and a role for HLA-DM in stabilizing empty HLA-DR molecules

of special interest. This study and [68] clarity the forms of MHC class II/invariant chain complexes capable of interacting with HLA-DM. They indicate that while the most abundant complex is with MHC class II/class II-associated invariant peptide, HLA-DM also associates with MHC class II/p 10 (see Figure 1) and empty MHC class II dimers. HLA-DM appears capable of promoting dissociation of the p 10 form of invariant chain from MHC class II in vitro, but clearly the preferred substrate is MHC class II/class II-associated invariant peptide, which is rapidly dissociated in the presence of HLA-DM
