Expression analysis in full locus transgenics in which a marked beta gene was placed at different positions in the locus. Primary transcript in situ hybridization data show that the LCR proximal β-gene has a distinct advantage over the distal β-gene in cis and results in the proximal gene being transcribed much more frequently than the distal gene. The mRNA expression levels of the different genes are directly proportional to the percentages of active genes in the population, showing again that a gene is either fully on or off.

Silencing of human fetal globin expression is impaired in the absence of the adult β-globin gene activator protein EKLF
-
-/- background β-globin expression was absent, and γ-globin was increased approximately fivefold. Burst forming unit-erythroid colonies derived from these mice showed similar results and excluded anemic stress as the cause of increased γ-globin expression. These data are discussed within the context of EKLF,s key role in facilitating the probability of β globin over γ globin expression.

Cell 1996, 87:105-114. Thorough analysis of full-locus transgenic mice with small deletions of the individual HS of the LCR. The data show that deletion of a single HS results in mild to severe reductions in expression owing to position effects at the site of integration in the host genome. Two different position effect mechanisms were noted. Reduced expression in the case of position effect variegation is caused by stable silencing of the transgene in a clonal subpopulation of erythroid cells. Cell timing effect results in low-level transgene expression in all erythroid cells, but transcription of the transgene is switched on and off in response to cellular signals.

Proc Natl Acad Sci U S A 1997, 94:169-174. YAC transgenics with small deletions of either the ε silencer or the 3′ β enhancer show that expression of both ε and γ is disrupted in embryonic cells with silencer mutations and expression of both γ and β is disrupted in fetal cells with β-enhancer mutations. A number of possible mechanisms are discussed that could account for the observed effects, including the idea of separate chromatin domains.
