This paper shows conclusively that cells of the myeloid lineage harbour CMV and that permissiveness of such cells for CMV replication is critically dependent on the differentiation state of the cell. CMV is demonstrated to reactivate in macrophages on allogeneic stimulation in vitro. This experimental system can be used to identify mechanisms involved in the maintenance of CMV latency and to explore the cellular activation pathways that mediate CMV reactivation. Additionally, this cell culture system will provide a way to examine mechanisms of CMV immunological surveillance as well as therapeutic approaches to prevent viral reactivation.

HHV-8 was found in the bone marrow dendritic cells of multiple myeloma patients but not in malignant plasma cells or bone marrow dendritic cells from normal individuals or patients with other malignancies. Viral IL-6, the human homologue of which is a growth factor for myeloma, was found to be transcribed in the myeloma bone marrow dendritic cells. HHV-8 may be required for the development and perpetuation of myeloma cells.
