Arterioscler Thromb Vasc Biol 1997; 17:475-482. This work clearly demonstrates that fibroblast growth factor-2 stimulates collagen degradation by inducing MMP-1 and reducing TIMP-2 production as well as inhibiting collagen synthesis. The authors suggest that growth factors may be critical in coordinating the balance between ECM synthesis and degradation.

It was observed that thrombin causes activation of MMP-2 and increased production of MMP-1 and MMP-3 through G-protein coupled thrombin receptor signalling in endothelial cells in vitro, suggesting that thrombin may be involved in tissue remodelling and angiogenesis.

This review summarizes this groups findings concerning a genetic variation in the human stromelysin promoter. The authors found that the 6A allele has a lower promoter reactivity and is associated with the progression of atherosclerosis. Therefore, they suggest that this allele favours matrix deposition and lesion development.
