Cell 1997, 89:755-764. Cbfa1-deficient mice completely lack bone formation and the maturation of osteoblasts and osteoclasts is blocked. Cbfa1 is preferentially expressed in osteoblasts and the markers of osteoblast differentiation are barely detectable in Cbfa1-deficient mice. These findings clearly indicate that Cbfa1 is an essential transcription factor for bone formation, especially osteoblast differentiation.

Cell 1997, 89:765-771. This study also shows the lack of bone formation in mice homozygously mutated in the Cbfa1 locus and presents in detail the abnormalities found in these heterozygously mutated mice. These abnormalities are similar to those of CCD. Cbfa1 was deleted in radiation-induced Ccd mutant mice. Cbfa1 expression in embryogenesis was examined using reporter β-galactosidase inserted in the Cbfa1 locus and Cbfa1 expression in the early stage (d10.5) was examined.

Osf2/Cbfa1: A transcriptional activator of osteoblast differentiation

3 suppresses Cbfa1 expression in primary osteoblasts. Transfection of the Cbfa1 expression vector induces osteoblastic markers, and antisense oligonucleotide suppresses type I collagen expression in vitro. These data suggest that Cbfa1 is a specific transcription factor for osteoblast differentiation.

Antisense oligonucleotides of Cbfa1 inhibit the production of alkaline phosphatase and osteocalcin, and decrease the formation of mineralized nodules - indicating that Cbfa1 is a specific transcription factor for osteoblast differentiation. Overexpression of all three runt-domain genes (Cbfa-1, -2, and -3) induces osteocalcin expression - supporting the importance of a runt-homology-domain protein in regulating the expression of a bone-specific gene.

J Biol Chem 1998, 273:4892-4896. Glucocorticoid is found to suppress Cbfa1 and the TGF-β type I receptor. It is suggested that bone loss caused by glucocorticoid may be mediated in part by Cbfa1 via the TGF-β type I receptor as the promoter for TGF-β type I receptor contains a Cbfa1-binding site. This paper indicated the importance of Cbfa1 in glucocorticoid-dependent bone loss.
