This paper determined whether renal disease aggregated in select families or was a sporadic complication in patients with SLE. Familial clustering of ESRD was observed in black patients with SLE who had nephritis, suggesting there is an inherited susceptibility to renal failure independent of the cause of ESRD.

Lupus 1996, 6:57-62. The authors found a significantly increased frequency of HLA-DR7 in SLE patients with antiphospholipid syndrome. Moreover, the distribution of HLA alleles in SLE patients resembles that found in their anticardiolipin antibody-positive first-degree relatives.

TNF microsatellite allele associations are not independent of HLA but may be important in the expression of certain clinical features of SLE.

In vitro apoptosis and expression of apoptosis-related molecules in lymphocytes from patients with systemic lupus erythematosus and other autoimmune diseases

Arthritis Rheum 1997, 40:306-317. The authors isolated lymphocytes from patients with SLE, rheumatoid arthritis, and other connective tissue diseases and measured the percentage of apoptosis after activation. Although the expression of Fas protein was increased in SLE lymphocytes, mRNA levels of Fas, FasL, and bcl-2 did not differ between SLE and control cells. The authors suggest that increased Fas and bcl-2 protein expression may not be specific for SLE.

J Clin Invest 1997, 99:725-731. The authors studied a candidate region with homology to a mouse SLE susceptibility locus on human chromosome 1q. Sibpair analysis in three ethnic groups showed evidence of linkage in a region on 1q41-42, suggesting that this candidate region contains an SLE susceptibility locus.
