A high resolution X-ray crystal structure of an unmodified hammerhead ribozyme is described in this article. Self-cleavage of crystals in the presence of added divalent metal ions was demonstrated and it was found that the significant conformational changes were limited to the active site.

In principle, a trans-cleaving hammerhead ribozyme can be assembled in several different constructs. More commonly, helices I and III are formed by substrate binding to give a catalytically active ribozyme. Alternatively, a substrate can also bind to form helices I and II. The latter construct was found to have different kinetics for cleavage compared to the I/III format.
