These authors mated transgenic mice lacking NPY with ob/ob mice. The resulting NPY- and leptin-deficient mice were markedly less obese than the ob/ob parental strain. The offspring also had lower serum glucose and insulin levels and were more fertile than the ob/ob parental strain.

This paper described the cloning of the NPY Y5 receptor which has pharmacological properties that closely match the pharmacological properties of the receptor that mediates the effects of NPY on feeding behavior, and appears to be the preferred target for anti-obesity drugs.

Nat Genet 1997, 15:269-272. This paper describes a novel uncoupling protein (UCP2) with homology to UCP1 but a much more widespread tissue distribution, suggesting the potential for regulation of energy dissipation in tissues other than the relatively nonabundant BAT.
