The authors of this paper used a variety of biochemical and molecular approaches to show that anchorage-dependent cyclin D1 expression is causal for pRb phosphorylation in NIH-3T3 cells. It also reports that cyclin E-cdk2 activity (measured in vitro] is anchorage-dependent in NIH-3T3 cells and that this effect is associated with changes in the expression of both p21 and p27. Results obtained with normal human fibroblasts are similar to those found with NIH-3T3 cells. But NRK cells, another anchorage-dependent cell line, seem to have lost the anchorage requirements for expression of cyclin D1, phosphorylation of pRb and activation of cyclin E-cdk2. These latter results support the idea that anchorage-dependent growth of NRK cells is controlled by a pRb-independent mechanism that allows for transcription of the cyclin A gene. The basis for the different cyclin E-cdk2 results obtained in this study is not yet clear.

The authors used NIH-3T3 cells to show that cyclin D and E kinase activities (measured in vitro) and pRb and p107 hyperphosphorylation (measured by gel shifts with whole cell lysates) are dependent upon cell anchorage. The absence of p107 phosphorylation prevents disruption of the E2F-p107 complex; the complex remains on the cyclin A promoter and cyclin A gene expression is blocked despite the presence of growth factors. Using in vitro assays, the authors also show that p27 degradation is blocked if cells are cultured in the absence of substratum, and they propose that this stabilization of p27 plays a role in the inhibition of cyclin D and cyclin E dependent kinases. The authors also showed that forced expression of cyclin D1 rescues cyclin A transcription and allows for S-phase entry in nonadherent 3T3 cells.

The effect was traced to increased expression of p21 and p27. Regulatory phosphorylation of cyclin E-cdk2 was also examined but seemed not to play a role in controlling kinase activity in response to substratum.

The authors show that Rb phosphorylation, cyclin E-cdk2 activity, and cyclin A expression are anchorage-dependent in PKC3-F4 and ER-1-2 cells. Overexpression of Ras allows for anchorage-independent expression of all three events in PKC3-F4 cells, but the cyclin A step remains anchorage-dependent in ER-1-2 cells. The study concludes that pRb phosphorylation is not sufficient to induce expression of the cyclin A gene and that transformation by Ras overrides several adhesion-dependent cyclin-cdk events.
