Promoter-proximal pausing by pol II and activator-dependent release of pol II from pausing is reconstituted for the first time in a cell-free system. Extended pausing by pol II downstream of the human hsp70 promoter is found to require the presence of nucleosomes on the DNA template. Release of pol II from pausing is shown to depend on the transcriptional activation domain of HSF1 and to require ATP and a fraction containing the SWI/ SNF complex, suggesting that chromatin remodelling may be a major step in the activator-dependent regulation of elongation. The assays described in this paper provide an excellent starting point for the biochemical characterization of promoter-proximal pausing and the mechanisms by which activators regulate this process.

These papers by Yankulov et al. [8] and Krumm et al. [9*] provide direct demonstrations that DNA-binding transactivators can promote elongation by pol II. These findings have forced a reappraisal of the roles of DNA-binding transactivators in gene expression and they have opened up a new area of research on elongation by pol II.

In this elegant study, P-TEFb is shown to be a CTD kinase and the CTD is shown to be required for P-TEFb-dependent stimulation of pol II elongation in vitro. These findings provide the first direct biochemical evidence that elongation by pol II can be controlled by a CTD kinase and they provide a working model for the mechanism of DRB-induced arrest, a long-standing issue in studies of transcriptioral regulation.

The authors provide an explanation for the observation that the AdML major ate arrest site blocks transcription by pol II in vitro when located downstream of the AdML promoter but not of the mouse β-globin promoter. Hybridization of an anti-arrest element at the 5́ end of the β-globin transcript or small anti-sense RNA or DNA oligonucleotides to the nascent transcript upstream of the arrest site is shown to block arrest, leading to the proposal that hybridization of the anti-arrest element to the nascent transcript blocks a pol II conformational change that occurs upon arrest and that involves retraction of the nascent transcript into the interior of pol II.

Interaction of TFIIF with elongating pol II decreases the likelihood that the enzyme will suffer arrest upon encountering an SII-responsive arrest site. Thus SII and TFIIF appear to control elongation by pol II in different yet complementary ways: whereas TFIIF decreases the duration of pausing by pol II and protects the elongation complex from arrest, SII reactivates elongation complexes that have arrested.

The VHL protein is shown to bind specifically to the Elongin BC complex both in vitro and in vivo. In addition, several VHL mutants found in tumors are shown to be defective in their abilities to bind to the Elongin BC complex, supporting a role for the VHL-Elongin BC interaction in VHL tumor-suppressor activity.

Proc Natl Acad Sci USA 1996, 93:1770-1775. This paper presents evidence that the subcellular localization of the VHL protein is regulated by cell-cell contact. The VHL protein is shown to reside in the cytoplasm of confluent cultured cells and in the nucleus of sparse cells. These findings suggest that the VHL protein may perform its tumor- suppression function as a component of a signalling pathway regulated by cell density.

[44*] report that expression of the VEGF gene is mis-regulated and constitutive in cells lacking a functional VHL protein. In addition, Iliopoulos et al. [42*] and Gnarra et al. [43*] propose that the VHL protein may have a role in regulating VEGF mRNA turnover, on the basis of findings indicating that, first, in transient transfection assays, reporter gene expression controlled by VEGF upstream sequences is similar in cells containing wild-type or mutant VHL genes; second, that pol II density on the endogenous VEGF gene, as measured by nuclear run-on assays, is similar in cells containing wild-type or mutant VHL genes; and third, that VEGF mRNA stability appears to be increased in one of several VHL mutant cell lines examined.

The authors of this paper describe the identification of a novel pol II elongation factor and demonstrate that it is encoded by the human ELL gene, which is a frequent target for translocations in acute myeloid leukemia. Taken together with evidence for a possible role of Elongin in the VHL familial cancer predisposition syndrome, this study provides the second potential link between transcription elongation and oncogenesis.
