Requirement for CD40 ligand in costimulation induction, T cell activation and experimental allergic encephalomyelitis

of outstanding interest. This study shows that CD154 is required for T cell priming and provides evidence for the regulation in vivo of the induction of co-stimulation by the CD40-CD154 system. Data show that T cells in CD154-deficient mice fail to upregulate co-stimulatory activity in draining lymph nodes upon antigen challenge. In addition, self antigen reactive TCR transgenic T cells fail to be activated and are unable to cause autoimmunity in these mice. See also [6].

CD40L-deficient mice show deficits in antiviral immunity and have an impaired memory CD8+ CTL response
-
of special interest. This study, along with [44,45], shows that CD40-CD154 interactions are critical for antiviral humoral responses. These interactions, however, are not required for the development of primary antiviral CTL responses. Furthermore, it was demonstrated that CD154 plays a critical role in the development of CTL memory responses.
