Science 1996, 274:1527-1531. Starting from the principle that neurotransmission mediated by 5-HT (serotonin) contributes to many physiologic functions; cognition; and emotional states, including anxiety, the authors thought that the 5-HT transporter might play a role in the fine tuning of serotoninergic transmission in the brain. Expression of that gene had been shown to occur in regions of the brain, such as the limbic system, that are known to play a role in emotional regulation. The authors identified two alleles of the transporter, one of which was transcribed less efficiently than the other and therefore made less protein, leading to reduced serotonin uptake. Reduced uptake, and hence increased serotoninergic transmission, is anxiogenic in animal models as well as humans. Gene-trait associations were found not only in a general population sample, where population stratification could have accounted for the association, but also in a separate study of sibling pairs with discordant genotypes.

The possibility that novelty-seeking behaviors could be related to dopamine activity is theoretically based on the results of animal studies (exploratory approach behavior), the effects of dopamine agonists in humans (the stimulation of euphoria, for example), and observations that persons with Parkinson's disease are both dopamine deficient and low in novelty seeking. Twin studies had shown that key temperament dimensions, including Novelty Seeking, Harm Avoidance, Reward Dependence, and Persistence, are moderately heritable (40%-60%).

The association between long alleles of exon III and personality traits related to novelty seeking, reported by Ebstein et al. [25], were confirmed. The association appears to be a result of genetic transmission rather than population stratification.

Evidence is presented to show that polymorphisms of three different dopaminergic genes show a significant correlation with behavioral features and that each gene, when examined individually, showed a significant but modest effect upon many of the same behaviors. The conclusion is reached that the Tourette's syndrome has a polygenic mode of inheritance and the exact mode of clinical presentation depends on the relative contributions of these modes in any one person.

Certain genes are expressed either from the maternal or the paternal genome as a consequence of imprinting; this article reports extraordinary findings on the cumulative effects of imprinted genes on brain development in the mouse, using a chimeric embryo as the experimental model. Androgenetic (duplicated paternal genome) cells contributed substantially to the hypothalamic structures but not to the cortex. Gynogenetic (duplicated maternal genome) cells contributed primarily to the cortex, striatum, and hippocampus but not to hypothalamic structures.

Evidence is presented for the existence of an imprinted locus on the X chromosome that influences the development of social-cognitive skills and associated higher cognitive functions. The presence of this locus may account, in part, for the evolution of sex differences in social perception and behavior. Its unusual mode of inheritance may explain why males in general are substantially more vulnerable than females to neurodevelopmental disorders associated with executive skill (prefrontal cortical) deficits, such as autism.

The author points out that we need to consider the relations between different levels of analysis to come to an integrated explanation of neurodevelopmental disorders. Genetic progress would be facilitated by refinement of the neurobehavioral phenotype. The first step in finding the genetic basis of complex disease will often be analysis of the phenotype itself. Usually, the phenotype studied in linkage or association studies will be relatively coarse from a cognitive perspective. It is therefore likely to be influenced by numerous underlying cognitive and genetic processes.

Meticulous research into the cognitive processes that are disrupted in dyslexia has shown that certain dyslexics have a deficit in phonologic decoding and consequently have slower and less accurate single-word recognition. They are also lacking in phonologic awareness and have difficulty segmenting phonemes to read an alphabetic language, such as English. The authors claim that cognitively dissociable components of the reading process are linked to separate genes and that the mapping from genes to aspects of cognition may be quite close. In other words, the hypothetical gene on chromosome 6 influences the development of phonologic awareness, but not single-word reading. On the other hand, the linkage on chromosome 15 (rather weaker, it must be said) is to the skill of single-word reading. There is, arguably, a dissociation between phonologic awareness and decoding and between phonologic awareness and single-word reading in the sample studied. Yet phonologic decoding and single-word reading were strongly correlated with one another.

These authors showed that submicroscopic chromosome 7q11.23 deletions associated with a partial Williams syndrome phenotype [59] cosegregated with poor visuospatial skills in two families. They hypothesized that the Williams syndrome was a contiguous gene deletion syndrome in which a typical cognitive profile includes a weakness in pattern construction and a strength in digit recall relative to performance on other nonverbal subtests. They tested the specific hypothesis that hemizygosity of a gene encoding a protein kinase that lay immediately 3′ of the elastin gene ELN could be responsible for the cognitive profile, because mouse models with protein kinase gene disruption had already been shown to have impaired visuospatial skills. LIMK1 (which is expressed widely in the cerebral cortex) mapped close to ELN locus and, in persons with the partial syndrome, the deletion extended to include that locus as well. Persons with the full Williams syndrome phenotype had much more extensive deletions that included both of the above genes, but LIMK1 was not deleted in persons with autosomal dominant supravalvular aortic stenosis only, in whom only the ELN gene was deleted (this gene is not expressed in the brain).

Adults with this syndrome are likely to develop a variety of serious mental illnesses, especially bipolar affective disorder, which often present initially during adolescence. A fascinating microdeletion syndrome, the further study of which could lead to the discovery of candidate genes for a major psychiatric disorder.
