This paper demonstrates that large errors in the equilibrium constants (here up to a factor of 500) can be introduced by their determination via kinetic experiments and kinetic analysis. In this study, even the ranking of different mobile reactants was affected. It is cautioned about rebinding and bivalency effects. A more reliable competition approach is described. It uses the change of the observed rate constant in the association phase upon addition of, for example, a soluble form of the immobilized binding sites as a competitor.

In this theoretically oriented paper, it is shown that traditional approaches to account for mass transport influence [8] can lead to concentration-dependent errors in the obtained apparent rate constants. A compartment model to quantify mass transport influence can be used only if restricted to the approach to equilibrium and to the dissociation phase. Analytical corrections for mass transport influence are derived, including a differential equation for combined dissociation and rebinding.

Whereas the simple 1:1 pseudo-first order model does not fit, several other models can globally fit experimental data comparably well. The importance of experimental controls is emphasized.

For cuvette-based biosensors, an equilibrium titration technique is proposed. An empirical, purely operational interpretation of binding rate constants can be used (in the absence of a model for kinetics) to measure the thermodynamic equilibrium constant in a solution competition approach.
