Extension of the mouse Npc1 cDNA sequence (GenBank accession number AF003348) was obtained by three sequential rapid amplification of cDNA ends (RACE) reactions from Marathon-Ready brain and liver libraries (Clontech). Section 1 was obtained by 3′ RACE polymerase chain reaction (PCR) amplification with primer mp25-1F (5′-CGTTCTGTCATTCAGTGTTGCGG-3′) with sequence from clone MW83C06. Sections 2 and 3 were obtained in sequential 5′ RACE reactions: section 2, mp25-2R (5′-CAGTCAAAGTAGTCATCGATCCAG-3′) followed by nested primer mp25-4R (5′-CTCAGCTGCGTTAAATATCTGC-3′); section 3, mp25-ZR (5′-CAACACAAGCCACGGGAACACC-3′) followed by nested primer mp25-LR (5′-GACCAGAGCTCTACAGGATTG-3′). The complete sequence of murine clone AA002656 has been submitted to GenBank (accession number AF003352).
