EMBO J 1996, 15:6132-6143. A very nice use of reverse genetic knockout worms. Here and in [22], the authors made targeted deletions in C. elegans homologs of MRP and MDR type multidrug resistance genes and initiated a functional study. They found that both MRP and MDR proteins give protection from toxic compounds (here being heavy metals) and that animals missing one gene of each type are hypersensitive, suggesting that they somehow act together.

Proc Natl Acad Sci USA 1996, 93:14940-14944. The use of C. elegans as a system for studying the function of human presenilins, genes that can be mutated to cause Alzheimer's disease. In previous work [32], the authors showed that the gene sel-12 encodes a presenilin homolog. Here, they found that intact presenilins can substitute for SEL-12 in vivo whereas the Alzheimer's disease causing mutant genes tested all have reduced activity. This provides an assay system and a starting point for further functional studies of human presenilins.

The authors of this paper show that ces-2, a cell-death-specification gene, encodes a basic region leucine-zipper transcription factor. Intriguingly, ces-2 is homologous to HLF, an oncogenic form of which inhibits programmed cell death in mammalian cells. The authors suggest that the normal role of HLF may be like that of ces-2, to negatively regulate a cell survival gene and that the oncogenic form has positive rather than negative activity.

Further molecular study of ced-4, a genetically identified cell-death gene. The authors describe a new alternatively spliced form of ced-4. The previously identified ced-4 transcript (ced-4S) can induce programmed cell death whereas the newly identified one (ced-4L) protects cells from programmed cell death. They propose that the balance between the opposing ced-4 functions determines whether a cell either lives or dies. A mammalian ced-4-like gene has not yet been found but is predicted to exist.

A beautiful demonstration of the power of selecting mutants with altered response to a drug. The authors isolated a large number of mutants resistant to inhibitors of cholinesterase. Many of the genes identified were found to be generally involved in synaptic transmission, whereas others act specifically in the acetylcholine pathway.
