Free phosphate ion, released from a substrate phosphopeptide (KRpTIRR), was quantified by colorimetric analysis (630 nm) using the Malachite Green method. p-Nitrophenyl phosphate was used as a substrate for the PP2B assay. Reaction was measured by the absorption of a hydrolyzed product, p-nitrophenol (415 nm).
