An in vitro assay for the inhibition of microtubule polymerization identified Op18/stathmin as a microlubule catastrophe factor. Op18/stathmin appears to exact its effect by binding to tubulin dimers. Op18/stathmin had previously been identified as an oncoprotein.

    • Walczak CE, Mitchison T, Desai A: XKCM1: a Xenopus kinesin-related protein that regulates microtubule dynamics during mitotic spindle assembly. Cell 1996, 84:37-47. XKCM1 was the first catastrophe factor to be identified using a cloning approach. Depletion of the protein in Xenopus egg extracts results in long microtubules because of a decrease in the catastrophe frequency. It is localized onto spindle poles and kinetochores and may play important roles during chromosome oscillation in prometaphase and segregation in anaphase.

    • Li Y, Benezra R: Identification of a human mitotic checkpoint gene: hsMAD2. Science 1996, 274:246-248. This paper describes the cloning of human MAD2 and the kinetochore localization of the protein. Interestingly, MAD2 localizes only to kinetochores that are not attached to microtubules. Antibody electroporation shows MAD2 to be necessary for mitotic arrest in response to spindle disruption.

Curr Biol 1996, 6:1609-1620. cdc55 mutant S. cerevisiae are able to inactivate mitosis-promoting factor and exit mitosis even in the presence of spindle damage. This escape is achieved via inhibitory phosphorylation on Cdc28, suggesting that Cdc55 is required to maintain phosphatase activity in order to maintain a mitotic arrest.

    • Genetic instability in colorectal cancers
    • Lengauer C, Kinzler KW, Vogelstein B: Genetic instability in colorectal cancers. Nature 1997, 366:623-627. This paper shows that several aneuploid colorectal cancer cell lines exhibit a continuous chromosome gain and loss, suggesting that these cells have an inherent defect in chromosome segregation. It is demonstrated that mere aneuploidy does not necessarily result in karyotypic instability, suggesting that the observed defect is due to a genetic mutation.

    • Hardwick KG, Weiss E, Luca FC, Winey M, Murray AW: Activation of the budding yeast spindle assembly checkpoint without mitotic spindle disruption. Science 1996, 273:953-956. Overexpression of Mps1 causes cell cycle arrest, even in the presence of a normal mitotic spindle. Overexpression also induces phosphorylation of Mad1, a hallmark of the activation of the spindle assembly checkpoint.
