The authors carefully analyze the functional properties of anti-HEL Tg B cells in mice that express sHEL when the anti-HEL Tg B cells either constitute a small minority or the vast majority of the recirculating B cell pool, They show that the increased survival of Tg B cells in the latter case can be attributed to decreased levels of circulating sHEL. This report damonstrates how relatively subtle changes in self-antigen expression can have significant repercussions on tolerance induction.

Concurranl activation through CD40 and the slgM receptor also results in increased Fas expression but these cells are resistant to FasL-expressirig T cells. These data suggest that, in vivo, Fas-FasL interactions are a critical faclor in the elimination B cells that have failed to receive a complete signal through their antigen-specific receptor.

By fusing B cells from phosphocholine immunized mice to bcl-2 transfected myeloma cells, the authors were able to isolate a remarkably high frequency of DMA-reactive hybridomas. Comparable numbers of DNA-reactiva hybridomas were not obtained with a conventional fusion partner. These results indicate that autoreactive B cells can be generated by somatic mutation during the response to conventional antigen, but such B cells normal die unless rescued by the bcl-2 fusion partner.

Proc Natl Acad Sci USA 1995, 92:11628-11632. B6/+ and B6/lpr mice were shown to respond comparably to hapten conjugated protein. The number of antibody-forming cells and memory B cells generated in the two strains were similar and sequence analysis of germinal center B cells revealed no evidence for the increased accumulation of low-affinity hapten-bindrng cells. These data indicate that the Ipr mutation does not significantly alter germinal center responses.
