The first X-ray structure of a human SDR Is presented, with direct bearing on carcinogenesis. This enzyme catalyzes the reduction of the weak estrogen, estrone, to the most potent, estradiol. Excess estradiol is thought to stimulate the so-called 'hormone dependent' breast tumors.

Common themes in redox chemistry emerge from the X-ray structure of oilseed rape (Brassies napus) enoyl acyl carrier protein reductase
-
Rafferty JB, Simon JW, Baldock C, Artymiuk PJ, Baker PJ, Stuitje AR, Slabas AR, Rice DW: Common themes in redox chemistry emerge from the X-ray structure of oilseed rape (Brassies napus) enoyl acyl carrier protein reductase. Structure 1 995, 3:927-938. The X-ray determination of enoyl acyl carrier protein reductase showed for the first time that a member of the SDR family could function without the YXXXK sequence. In this protein an analogous position of the catalytic residues tyrosine and lipase, relative to the colactor location, is achieved in a novel fashion.

Crystal structure and function of the isoniazid target of Mycobacisrium tuberculosis
-
Dessen A, Ouomard A, Blanchard JS, Jacobs WR Jr, Sacchettim C: Crystal structure and function of the isoniazid target of Mycobacisrium tuberculosis. Science 1995, 267:30-41. This paper presents an X ray structure which led to the realization that an enzyme critical to successful treatment of tuberculosis is a member of the SDR family.
