    • M. The crystallographic analysis reveals that each subunit ol this tetrameric protein is formed by three domains linked by flexible hinges. The orientation of the domains is different in the crystallographically independent subunits. indicating that interdomam flexibility is likaly to play a role in the allosteric regulation.

Refinement of the GroEL structure shows that the apical domain is able to move as a rigid body adopting different orientations in the crystallographically independent subunits. This paper is also innovative from a methodological point of view because it originates from the application ol an improved crystallographic refinement technique based on torsion-angle dynamics.

    • Unwin N: Acetylcholine receptor channel imaged in the open state. Nature 1995,373:37-43. An elegant electron microscopy study of the acatylcholine receptor activated by < 5 ms mixing with droplets containing acetylcholine. Activator binding on the extracellular portion of the protein induces a change in the configuration of the helices forming the channel wall in the membrane-spanning region. The activation mechanism derived from this analysis is likely to be of general validity for other ion channels.

    • Hunt JF, Weaver AJ, Landry SJ, Giarasch L, Deisenhofer J: The crystal structure of the GroES co-chaperonin at 2.8 Å resolution. Nature 1996, 379:37-45. Two papers [27,28'] show that the GroES heplamer forms a dome which binds at the top of the GroEL cylinder. The strength of the interaction is different in the ATP-bound and ADP-bound forms of GroEL. Thus, GroES exemplifies the case ol a protein acting as all oat eric effector.
