The continuation of a functional screen for molecules rescuing axial development in ventralized Xenopus embryos has led to the isolation of Xnr-3 cDNAs, which encode a protein related to mouse nodal (see [10,11]). Xnr-3 does not induce mesoderm but causes embryonic cells to undergo gastrulation-like movements. The reported activities and expresson pattern indicate a role for Xnr-3, as with mouse nodal, in regulating morphogenesis during gastrulation.

Ectopic expression of mouse nodal in Xenopus embryos has revealed a very potent dose-dependent mesoderm-inducing activity that is consistent with the embryonic phenotype caused by inactivation of this gene in mouse (see [10,11]). cDNAs for the Xenopus nodal-related genes Xnr-1 and Xnr-2 were shown to encode probable functional homologs of nodal. Various complementary bioassays strongly suggest that a signaling pathway involving nodal/nodal-related genes is a conserved element in the mesoderm differentiation associated with vertebrate gastrulation and patterning.

Homozygous Bmp7 mutants die at birth because of renal failure. Kidney development appears normal until -11.5 days and then ceases, suggesting that BMP7 is needed for the proliferation and differentiation of the metanephric mesenchyme. Defects are also seen in eye development and most embryos show poldactyly, consistent with a role for BMP7 in eye and limb patterning.

In this report, the authors detail the cloning of schnurri, which is found to encode a zinc-finger protein related to the mammalian family of transcription factors. Thus schnurri may be a critical downstream component of the intracellular DPP signal transduction pathway.

The Drosophila homolog of vertebrate BMP4, DPP, promotes ventral cell fates in Xenopus. Injection of sog mRNA counteracts the ventralizing effects of dpp. These experiments support the idea that the SOG and CHORDIN proteins play similar roles in antagonizing the activity of either DPP or BMP4.

In vitro assays using intermediate neural plate and markers for dorsal and ventral cell types, demonstrate that exogenous BMP4 and BMP7 counteract the ventralizing activity of the notochord and Shh.

By reconstructing serial sections after in situ hybridization at different stages, the authors show that these genes are expressed in nested domains of the epithelium in the region of the enamel knot. Bromodeoxyuridine labeling reveals that cell proliferation is reduced in the enamel knot, which may be acting as a signaling centre during tooth morphogenesis.

Overexpression of Bmp4 in the epithelium during lung development using the SP-C enhancer/promoter leads to severe lung abnormalities. Proliferation of the endoderm is inhibited, whereas that of the mesenchyme is stimulated.
