1. Couch FJ, Weber BL, and the Breast Cancer Information Core: Mutations and polymorphisms in the familial early onset breast cancer (BRCA1) gene. Hum Mutat 1996, 8:8-18. This paper is a review of BRCA1 mutation analysis and a complete listing of all BRCA1 mutations available at the time of publication. A number of the mutations described have not been reported elsewhere. In addition, there is a thorough description of the various types of mutations common to all genes.
2. Langston AA, Malone KE, Thompson JD, Daling JR, Ostrander EA: BRCA1 mutations in a population-based sample of young women with breast cancer. N Engl J Med 1996, 34:137-142. This study describes the incidence of BRCA1 mutations in a sample of women diagnosed with breast cancer before the age of 35. The figures generated may be useful in estimating the likelihood of finding mutations in individual patients.
3. FitzGerald MG, MacDonald DJ, Krainer M, Hoover I, O'Neil E, Unsal H: Germ-line BRCA1 mutations in Jewish and non-Jewish women with early onset breast cancer. N Engl J Med 1996, 334:143-149. Like the paper by Langston et al. (N Engl J Med 1996, 34:137-142), this group estimated the incidence of BRCA1 mutations in a sample of women diagnosed with breast cancer before the age of 30 and presents data similar to the Langston et al. (N Engl J Med 1996, 34:137-142) manuscript. In addition, the sample set assembled by these authors allow calculation of the incidence of BRCA1 mutations in young Ashkenazi Jewish women.

5. Struewing JP, Abeliovich D, Peretz T, Avishai N, Kaback MM, Collins FS, Brody LC: The carrier frequency of the BRCA1 185delAG mutation is approximately 1 percent in Ashkenazi Jewish individuals. Nature Genet 1995, 11:198-200. This manuscript is the first estimation of the 185delAG BRCA1 mutation rate in Ashkenazi Jewish individuals unselected with regard to breast cancer. The data, supported by ongoing work from this group and others, suggests that 1% of all Ashkenazi carry this specific breast cancer susceptibility mutation, a surprisingly high percentage.

This paper describes the construction of the first BRCA1 knockout mouse that has no functional copies of BRCA1 in its genome. The investigators demonstrate that the absence of functional BRCA1 results in an embroyonic lethal defect with death in utero between days 10 and 13. This work is the first evidence, as might be expected, that BRCA1 function encompasses more than suppression of breast and ovarian cancer.

This paper provides a good overview of cell adhesion systems in normal and malignant breast tissue. Although only 11 cases were analyzed, the investigation of a number of markers in the same tissue specimens provides important insights into differences between in the composition of adherens junctions in normal and malignant tissues.

This study suggests that, in aggressive breast carcinomas, when E-cadherin is downregulated, P-cadherin may mediate the formation and maintenance of tumor cell nests.

30. Siitonen SM, Kokoene JT, Helin HJ, Rantala IS, Holli KA, Isola JJ: Reduced E-cadherin expression is associated with invasiveness and unfavorable prognosis in breast cancer. Am J Clin Pathol 1996, 105:394-402. This study investigated the relationship between reduced E-cadherin staining and other clinicopathologic features. Although positive associations were largely consistent with earlier studies, this was the first study to associate E-cadherin staining with reduced disease-free survival. Although the multivariate analysis revealed borderline statistical significance (n = 109; P = 0.072), these findings should be validated in series of larger sample size and with longer follow-up.

Cathepsin D immunostaining was performed on 2524 samples. Clinical follow-up data was available for 489 patients. High cathepsin D staining was associated with the presence of axillary metastases and was significantly predictive of shortened relapse-free and overall survival.
