Nature Genet 1995, 11:198-200. This manuscript is the first estimation of the 185delAG BRCA1 mutation rate in Ashkenazi Jewish individuals unselected with regard to breast cancer. The data, supported by ongoing work from this group and others, suggests that 1% of all Ashkenazi carry this specific breast cancer susceptibility mutation, a surprisingly high percentage.

This paper describes the construction of the first BRCA1 knockout mouse that has no functional copies of BRCA1 in its genome. The investigators demonstrate that the absence of functional BRCA1 results in an embroyonic lethal defect with death in utero between days 10 and 13. This work is the first evidence, as might be expected, that BRCA1 function encompasses more than suppression of breast and ovarian cancer.

Although positive associations were largely consistent with earlier studies, this was the first study to associate E-cadherin staining with reduced disease-free survival. Although the multivariate analysis revealed borderline statistical significance (n = 109; P = 0.072), these findings should be validated in series of larger sample size and with longer follow-up.

Eur J Cancer 1995, 31A:671-677. This paper reports on the results of an 11 -year, five-center study of the relationship between cathepsin D and known prognostic factors in breast cancer. Cathepsin D immunostaining was performed on 2524 samples. Clinical follow-up data was available for 489 patients. High cathepsin D staining was associated with the presence of axillary metastases and was significantly predictive of shortened relapse-free and overall survival.
