    • In studies that are not limited to the small sample sizes analyzed in this work, concentration detection limits for two-photon fluorescence analysis could be improved substantially using a variety of approaches. In one strategy, the submicrometer laser focal spot position could scanned with galvinometer-driven mirrors to probe the outlet of much larger inner diameter capillaries. Alternatively, the size of the laser spot could be increased by underfilling the back aperture of the high NA objective (producing a Gaussian focus). In this approach, the maximum size of the probe area (<2-μm diameter) is limited by the available laser power.

    • A 70-nm-diameter vesicle packed with 0.2 M neurotransmitter contains ∼30 000 molecules, ∼2-fold higher than the detection limit for CPP-glycine. The components of such a vesicle, originally contained in a volume of ∼0.2 aL, would diffuse longitudinally and radially in a 620-nm-i.d. capillary to a volume of ∼80 fL in 200 s. The resulting concentration at the detection site thus would be ∼600 nM.

    • Fluorescence detection has been used to measure the contents of individual secretory granules from other cell types, in which the amount of material is 10-100-fold greater than that contained in neuronal vesicles.
