    • Flat-bottom 96-well microtiter plates (Corning Costar, Corning, NY) were coated overnight at 4°C with 100 μL/well of whole bacterial lysates of C. perfringens or C. septicum (10 μg/mL) in 0.1 M carbonate buffer, pH 9.6. Crude soluble clostridial antigens were prepared from C. perfringens and C. septicum cultures using a previously published protocol [Kulkarni, R. R., V. R. Parreira, S. Sharif, and J. F. Prescott. 2008. Oral immunization of broiler chickens against necrotic enteritis with an attenuated Salmonella vaccine vector expressing Clostridium perfringens antigens. Vaccine 26:4194-4203]. Plates were washed 3 times with PBS containing 0.05% Tween 20 (PBS-T), blocked with 200 μL/well of PBS containing 1% BSA for 1 h at room temperature, washed with PBS-T, and incubated with 100μL/well of appropriately diluted serum samples for 2 h at room temperature. After washing with PBS-T, horseradish peroxidase-conjugated goat anti-chicken IgG antibody (1:5,000; Sigma, St. Louis, MO) was added, and bound antibodies were detected at 450 nm after reaction with 0.01% tetramethylbenzidine (Sigma) in 0.05 M phosphate-citrate buffer, pH 5.0, using a microplate reader (Bio-Rad, Richmond, CA).
