The isocratic mobile phase, which consisted of methanol (HPLC grade, Fisher Scientific, Pittsburgh, PA):water:ascorbic acid (96:3:1), was maintained at a flow rate of 1.0 mL/min (CBM-10A), and automated injections (SIL-10A XL) of 50 μL were maintained.

HPLC analysis of carotenoids: The xanthophylls were measured by straight-phase HPLC using methanol:water:ascorbic acid (96:3:1) as the mobile phase at a flow rate of 1.5 mL/min. The solution was passed through a 0.45-μm membrane filter and immediately used for HPLC analysis. A C18 reversed-phase column (5 μm, 150 Å, 4.6 mm i.d. × 250 mm length; No. VS952505, Agela Technologies Inc., Newark, DE) and a high-performance guard column (5 μm; Vydac 201GD54T, Agela Technologies Inc.) were used. The column was chilled to 15°C with a water jacket (35 cm; Alltech, Deerfield, IL) and a circulating water chiller (RF-10; New Brunswick Scientific, Edison, NJ). Absorbance at 450 nm was monitored and peaks were integrated by using a UV/visible detector (SPD-10A) monitored at 450 nm. Millenium software (CLASS-LC10, Post Run Analysis, Shimadzu Corp., Tokyo, Japan) was used to process and integrate the peaks. Peak identification and quantification of sample concentrations were done by comparison with purified standards of xanthophylls (097k4075, Sigma, St. Louis, MO) and β-carotene (076k7051, Sigma). The respective retention times were 7.2 and 14.5 min, and the limit of detection for each carotenoid in our standards was 0.17 pmol. Three injections of each standard were made at different volumes before each sample set, and standard curves were generated using linear regression (JMP software, SAS Institute [31]).
