In vitro whole-organ imaging: 4D quantification of growing mouse limb buds

The authors demonstrate in vitro imaging of organogenesis in mammals by time-lapse OPT. Although the spatial resolution and the temporal sampling are moderate, the authors accomplish a very impressive imaging of the dynamics of expression patterns deep inside a developing mammal organ. The combination of advanced culturing methods with the unique imaging features of OPT fluorescence microscopy provides quantitative data that should be especially useful for computational modelling of organogenesis.

The quantitative analysis of microtubule dynamics in three dimensions provides data that allow the authors to formulate and test an extended kinetic model of microtubule dynamic instability.

The paper highlights the unique advantages of LSFM, which enable high-speed imaging of living embryos at good spatial resolution.
