    • PCL-PRC2 is needed to generate high levels of H3-K27 trimethylation at Polycomb target genes
    • This work provides biochemical and genetic evidence that PCL protein is required for generation of high levels of tri-methyl H3K27 in the chromatin of PcG target genes. This finding suggests that only a subset of E(Z)/ESC complexes that contain PCL represent PRC2 and are specifically involved in PcG silencing while more abundant variants of E(Z)/ESC complexes monomethylate and dimethylate H3K27 elsewhere in the genome.

    • A phosphorylated form of Mel-18 targets the Ring1B histone H2A ubiquitin ligase to chromatin
    • This careful study reveals the role of Mel-18 in direction of the ubiquitylation activity of Ring1B toward lysine 119 of histone H2A. It also shows that substrate-targeting function of Mel-18 is dependent on its prior phosphorylation at multiple residues, providing possible link for regulation of PRC1 activity by cell signaling pathways.
