PCL-PRC2 is needed to generate high levels of H3-K27 trimethylation at Polycomb target genes
-
This work provides biochemical and genetic evidence that PCL protein is required for generation of high levels of tri-methyl H3K27 in the chromatin of PcG target genes. This finding suggests that only a subset of E(Z)/ESC complexes that contain PCL represent PRC2 and are specifically involved in PcG silencing while more abundant variants of E(Z)/ESC complexes monomethylate and dimethylate H3K27 elsewhere in the genome.

A phosphorylated form of Mel-18 targets the Ring1B histone H2A ubiquitin ligase to chromatin
-
This careful study reveals the role of Mel-18 in direction of the ubiquitylation activity of Ring1B toward lysine 119 of histone H2A. It also shows that substrate-targeting function of Mel-18 is dependent on its prior phosphorylation at multiple residues, providing possible link for regulation of PRC1 activity by cell signaling pathways.

Genome-wide analysis of Polycomb targets in Drosophila melanogaster
-
Detailed genome-wide mapping of the distribution of PcG complexes on Drosophila chromosomes revealed localized binding of PcG proteins to the PREs embedded in broad domains of H3K27me3. PREs were found to be largely depleted of H3K27me3 probably owing to the general depletion of histone H3 from these elements.

Control of developmental regulators by Polycomb in human embryonic stem cells
-
The results of genome-wide mapping of PcG proteins in human and mouse embryonic stem cells reported in this and the following paper [37] provide a direct demonstration of the global role of PcG regulatory system in the control of cellular differentiation.

A bivalent chromatin structure marks key developmental genes in embryonic stem cells
-
This article contains the first notion of the surprising coexistence of H3K27me3 and H3K4me3 regions within chromatin of many PcG target genes in the embryonic stem cells and coins the term 'bivalent chromatin domain'.

Ring1-mediated ubiquitination of H2A restrains poised RNA polymerase II at bivalent genes in mouse ES cells
-
This thorough work shows that multiple processes are probably to be responsible for the generation of a balanced epigenetic state of PcG target genes in embryonic stem cells.
