The Pseudomonas syringae effector HopU1 is shown to encode a mono-ADP-ribosyltransferase enzyme that targets Arabidopsis RNA-binding proteins. The effector-mediated ADP-ribosylation of these host proteins at RNA recognition motifs is proposed to inhibit their post-transcriptional regulatory function. A knockout mutant of one of these Arabidopsis proteins (AtGRP7) showed enhanced susceptibility to virulent P. syringae and reduced callose formation when treated with a bacterial PAMP molecule (flg22) validating this protein as a pathogen target for the suppression of basal defense.

This study makes a mechanistic link between R gene-mediated resistance triggered by effectors and basal defense triggered by PAMP molecules. The barley powdery mildew resistance protein MLA10 is shown to function in the nucleus by associating with host WRKY transcription factors when the AVR10 mildew pathogen effector is present. These host transcription factors appear to function as repressors of PAMP-triggered basal defense. The AVR10-dependent association of the MLA10 protein with WRKY proteins is believed to suppress the function of these transcription factors resulting in defense pathway de-repression and subsequent disease resistance.

Pre- and postinvasion defenses both contribute to nonhost resistance in Arabidopsis

In conjunction with references [34,36] these papers describe three genes that form part of the Arabidopsis preinvasive, nonhost defense response to mildews and other pathogens. These genes, pen1-pen3, encode a syntaxin protein involved in membrane vesicle transport, a peroxisome localised glycosyl hydrolase and an ATP-binding cassette transporter, respectively. Mutation of each gene allows increased penetration frequency of Arabidopsis by phytopathogens for which this plant is a nonhost. Combining pen mutants with mutations in R gene signalling and basal defense pathways caused a dramatic increase in pathogen growth on this nonhost species suggesting distinct pre-invasive and post-invasive defense mechanisms.

Introduction of this gene into rice conferred resistance against the unrelated rice pathogen Xanthomonas oryzae pv. oryzicola for which maize is a nonhost. This is the first successful transfer of nonhost resistance between cereals.

Subcellular localisation and functions of the barley stem rust resistance receptor-like serine/threonine-specific protein kinase Rpg1

The RPG1 protein was demonstrated to function as a protein kinase capable of intramolecular autophosphorylation with this kinase activity being essential for resistance. Subcellular fractionation detected the protein in the cytosol, plasma membrane and intracellular membrane fractions.

The RPG1 protein is shown to be degraded in barley tissue infected with avirulent isolates of P. graminis but not virulent isolates. Degradation of the protein occurs via ubiquitination and the proteasome pathway.
