Javaid MK, Crozier SR, Harvey NC, et al. Maternal vitamin D status during pregnancy and childhood bone mass at age 9 years: a longitudinal study. Lancet 2006; 367:36-43. This longitudinal study shows that Reduced concentration of 25(OH)-vitamin D in mothers during late pregnancy was associated with reduced whole-body and lumbar-spine bone-mineral content in children at age 9 years. The authors suggest that vitamin D supplementation of pregnant women, especially during winter months, could lead to longlasting reductions in the risk of osteoporotic fracture in their children.

Feng JQ, Ward LM, Liu S, et al. Loss of DMP1 causes rickets and osteomalacia and identifies a role for osteocytes in mineral metabolism. Nat Genet 2006; 38:1310-1315. Mutational analyses have shown that 2 families with autosomal recessive hypophosphataemic rickets (ARHR) have mutations affecting the DMP-1 gene. DMP-1 null mice have high levels of circulating FGF-23 with a hypophosphataemic rickets phenotype. Defective DMP-1 produced by the osteocyte may result in elevated FGF-23 and ARHR.

Nat Genet 2006; 38:1248-1250. Mutation of the DMP-1 gene at 4p21 which encodes a noncollagenous bone matrix protein expressed in osteoblasts and osteocytes has been identified in subjects with autosomal recessive hypophosphataemic rickets. Two of these subjects have an elevated FGF23. Crucially, this may suggest that osteocyte/osteoblast DMP-1 may be an important upstream regulator of FGF23.
