The calvarial defect model of rats was prepared according to a method described below. The animals were anesthetized via an intraperitoneal injection with a cocktail of xylazine (Rompun®, 20 mg/ml, Bayer Korea Ltd., Korea) and ketamine hydrochloride (Ketara®, 100 mg/ml, Yuhan Corp, Korea) (1:4 volume mixture, dosage 0.1 ml/100 g body weight). After shaving the scalp hair, a longitudinal incision was made in the midline of cranium from nasal bone to the posterior nuchal line, and the periosteum was elevated to expose the surface of the parietal bone. Using a 8 mm surgical trephine (Ace Surgical supply, Brockton, USA) and small carbide round bur in low-rotational speed drill, calvarial critical size defect (CSD) with an 8 mm diameter was created. To excise exactly the defect out of the cranium for later evaluation, the long diameter of defect was located along the sagittal suture of calvaria. The cutting was done in unique thickness of calvarial depth under the magnification with operating microscope (Zeiss, Germany), and the defect was filled with the prepared implants.
