Immunohistochemical analysis revealed significantly increased collagen III expression in the uterosacral ligament of subjects with pelvic organ prolapse (P<0.001). Statistical analysis accounted for age and parity. Emphasized importance of examining collagen subtypes and their ratios instead of total collagen content.

Failure of elastic fiber homeostasis leads to pelvic floor disorder
-
Liu X, Zhao Yun. Pawlyk B, et al. Failure of elastic fiber homeostasis leads to pelvic floor disorder. Am J Pathol 2006; 168:519-528. Demonstrated LOXL1-deficient mice to be unable to maintain elastic fiber homeostasis after parturition, leading to pelvic organ prolapse and lower urinary tract dysfunction. Showed diminished expression of LOXL1 with aging. This study highlights the importance of elastin for maintaining structural and functional integrity of the pelvic floor. Underscores the role of genetic predisposition and advancing age in the etiology of pelvic floor dysfunction.

Increased expression of matrix metalloproteinase 2 in uterosacral ligaments is associated with pelvic organ prolapse
-
Gabriel B, Watermann D, Hancke K, et al. Increased expression of matrix metalloproteinase 2 in uterosacral ligaments is associated with pelvic organ prolapse. Int Urogynecol J Pelvic Floor Dysfunct 2005; 1-5. Demonstrated increased expression of MMP-2 in subjects with pelvic organ prolapse. Analysis accounted for age and parity. Underscored active remodeling of pelvic floor connective tissue associated with prolapse.

Estrogen increases collagen I and III mRNA expression in the pelvic support tissues of the rhesus macaque
-
Clark AL, Slayden OD, Hettrich K, Brenner RM. Estrogen increases collagen I and III mRNA expression in the pelvic support tissues of the rhesus macaque. Am J Obstet Gynecol 2005; 192:1523-1529. Measured the expression of collagen I and III mRNA in full-thickness anterior vaginal wall and lateral paravaginal attachment. Treatment with estradiol resulted in significant increase in mRNA expression of collagen I and III and in collagen content. The presence of this upregulation highlights the important role of estrogen in the balance between synthesis and degradation of collagen in the connective tissues of the pelvic floor.
